T-TAS®️ (Total Thrombus formation Analysis System), based on our unique technology using flow chamber system, can quantitatively measure thrombus formation process under physiological flow conditions.
ASSAY PRINCIPLE
The thrombus formation process is a biological function for repairing damaged blood vessels in the body. When a blood vessel is damaged, substances that activate thrombus formation, such as collagen and tissue thromboplastin, are exposed. Platelets in the bloodstream are activated by these substances and primary hemostasis progresses (platelet thrombus formation). In addition, tissue thromboplastin induces a cascade reaction of blood coagulation, which promotes secondary hemostasis (formation of white thrombus). Thus, blood flow conditions and collagen / tissue thromboplastins play important roles in thrombus formation.
The T-TAS system and dedicated microchip were developed to easily reproduce this thrombus formation reaction for ex vivo blood testing. The flow paths of microchips are coated with collagen or collagen and tissue thromboplastin, so that its structure imitates the environment of damaged blood vessels. The T-TAS system is a small device with a built-in pump and pressure sensor. When a dedicated microchip is inserted into the device and a blood sample is applied into the chip, the blood is pumped through the flow path. At this time, the flow rate is automatically set to an optimized value for each special chips so that in vivo environment is reproduced inside the chip. In addition, the pressure inside the flow path can be monitored as a time course by a pressure sensor, and it is possible to quantitatively observe how the pressure inside the flow path rises with the formation of thrombosis using the T-TAS system.
ASSAY RESULT
Changes of flow pressure are monitored by the pressure sensor located upstream in the chamber. Thrombus formation within the flow chamber increases flow resistance causing the pressure to increase. OST (Occlusion Start Time) is the lag time for the flow pressure to reach 10 kPa due to partial occlusion of the capillary. OT (Occlusion Time) is the lag time for the flow pressure to reach 60 kPa from baseline pressure. The AUC (Area Under the Curve) is the area under the flow pressure vs. time curve and is related to overall thrombus formation.

PL Chip
The PL-chip was specifically designed for the quantitative analysis of primary hemostatic ability, i.e. the overall platelet thrombus formation process, which involves platelet adhesion & aggregation, granule secretion, and thrombus growth.
ASSAY PRINCIPLE
*PL Chip for T-TAS®01: pre-coated with type I collagen (pig-derived) on its surface of the flow paths.
*BAPA tube for T-TAS®01: blood collection tubes containing spray-dried BAPA (benzylsulfonyl-d-arg-pro-4-amidinobenzylamide), an anticoagulant that inhibits factor Xa and thrombin.
*PL Chip Reservoir Set for T-TAS®01: contains accessories for use with the PL Chip for T-TAS 01.
When blood collected in the BAPA tube for T-TAS® 01 is applied into the PL chip’s flow path, platelets are activated by the effect of collagen and 1500 /s wall shear stress. Because of the BAPA anticoagulant, the secondary hemostasis is inhibited and resulting in the only primary hemostasis progress. As a result, platelet thrombi are formed, which blocks the inside of the flow path, and the pressure inside the flow path increases.

IMPORTANT NOTES
The T-TAS®01 PL chip is intended for use in the clinical laboratory for the analysis of the platelet thrombus formation process (primary hemostatic function) in patients aged 21 and older with a history of conditions associated with impaired primary hemostatic function or use of antiplatelet therapy.
Physicians should use their clinical judgment and experience when deciding how to diagnose and treat patients and in the use of the PL Chip in the treatment of the patient. Please refer to the PL Chip Package Insert and T-TAS®01 User’s Manual for full instructions on sample collection and handling, and all other test procedures.
AR Chip
AR chip is suitable for the assessment of whole-blood hemostasis (primary and secondary hemostatic functions).
ASSAY PRINCIPLE
*AR Chip: pre-coated with type I collagen (pig-derived) and tissue thromboplastin (rabbit-derived) on its surface of the flow paths.
*3.2% Citrate tube: blood collection tubes containing 3.2% sodium citrate
* CaCTI reagent for T-TAS®01: contains CTI(Corn-derived trypsin inhibitor, the inhibitor for FXII)and CaCl2
*AR Chip & HD Chip Reservoir Set for T-TAS®01: contains accessories for use with the AR Chip for T-TAS 01.
When citrated blood treated with CaCTI reagent is applied into the AR chip flow path, platelets are activated by the effect of collagen and 600 /s wall shear stress. Furthermore, blood coagulation pathway is activated by tissue thromboplastin and calcium ion. In this mechanism, intrinsic coagulation pathway is eliminated because of inhibition of FXII by the effect of CTI. In summary, white thrombi are formed as a result of primary and secondary (especially with extrinsic pathway) hemostasis and the pressure inside the flow path increases due to the occlusion of the flow paths.

HD Chip
HD Chip can measure whole-blood hemostasis (primary and secondary hemostatic functions) and work with specimen even with low platelet count.

*Anti-sedimentation reagent: contains DNDS (4,4'-Dinitrostilbene-2,2'-disulfonic Acid Disodium Salt) which inhibits blood cell precipitation during measurement
*AR Chip & HD Chip Reservoir Set: contains accessories for use with the HD Chip
When citrated blood treated with CaCTI reagent is applied into the HD chip’s flow path, platelets are activated by the effect of collagen and 1,200 /s wall shear stress. Furthermore, blood coagulation pathway is activated by tissue thromboplastin and calcium ion. In this mechanism, intrinsic coagulation pathway is eliminated because of inhibition of FXII by the effect of CTI. In summary, white thrombi are formed as a result of primary and secondary (especially with extrinsic pathway) hemostasis and the pressure inside the flow path increases due to the occlusion of the flow paths.
The depth of flow path in the HD chip is 50 µm, which is shallower than AR chip flow path. The wall shear stress which is higher than that of AR chip (HD: 1,200 /s, AR: 600 /s) makes it possible to measure thrombus formation process even in a blood sample with a low platelet count (10,000-90,000 /µL).
